家独行菜的组织培养与高频再生

摘 要:

以家独行菜萌发苗的茎和叶作为外植体, 利用含有多种不同植物生长调节物质及浓度配比的MS培养基分别诱导产生愈伤组织、丛生芽和再生根, 建立了组织培养和高频离体再生体系。结果表明: 最适外植体为苗龄14 d的家独行菜萌发苗茎段, 其诱导愈伤组织的最适培养基为MS+1.5 mg·L-1 2,4-D+0.5 mg·L-1 6-BA+0.1 mg·L-1 NAA, 诱导率可达95.4%; 诱导不定芽的最适培养基为MS+0.5 mg·L-1 2,4-D+1.5 mg·L-1 6-BA+0.2 mg·L-1 NAA+0.1 mg·L-1 TDZ, 诱导率可达89.9%; 在MS+0.6 mg·L-1 NAA的培养基中可获得最佳生根率, 大约有87.5%的再生苗生长出健康的根系。炼苗后将长势良好的再生苗进行移栽, 存活率可达90%。

Tissue culture and high-frequency regeneration of Lepidium sativum

Abstract:

The stems and leaves of Lepidium sativum seedlings were used as explants, MS medium compounding proportions of different plant growth regulators were studied on callus induction, adventitious bud differentiation, and root regeneration, thereafter an efficient plant-regeneration system was established by in vitro L. sativum culture. On MS medium containing 1.5 mg·L-1 2,4-D, 0.5 mg·L-1 6-BA and 0.1 mg·L-1 NAA, stems could be induced to product calli, and callus induction ratio reached to 95.4%. On MS medium containing 0.5 mg·L-1 2,4-D, 1.5 mg·L-1 6-BA, 0.2 mg·L-1 NAA and 0.1 mg·L-1 TDZ, adventitious buds could be regenerated from calli, and the differentiation ratio was 89.9%. Well-grown adventitious buds were inoculated on MS medium containing 0.6 mg·L-1 NAA, and 87.5% rooting frequency was obtained. Regenerated plantlets grew well after being transplanted to soil. Moreover, the survival ratio of plantlets reached 90%.
